BDNF boost
Growth signaling as such — and less of it is direct than the name suggests.
The idea
The title is a claim that has to be earned, so the correction goes first: only some of these four have direct BDNF evidence. What they actually share is neurotrophic signaling more broadly. Deep cognitive repair is about damage already done; this one is about growth signaling in a brain that is not necessarily damaged.
What's in it
| Compound | Role in this stack | Evidence |
|---|---|---|
| NSI-189 | Neurogenesis-linked small molecule; the item with the most human data, from depression trials. | Clinical, mixed — encouraging early phase II, larger study missed its primary endpoint |
| TAK-653 | AMPA positive allosteric modulator with minimal agonist activity: amplifies glutamatergic signaling only where it is already occurring, rather than generating its own. | Investigational; Phase 3 underway |
| P21 | Peptide fragment derived from ciliary neurotrophic factor, associated with neurogenesis in rodent work. | Preclinical; thin |
| Adamax | Adamantylated analogue in the Semax family, usually taken intranasally. | Investigational; thin |
Overlap & conflict
One overlap: the BDNF/TrkB lever is shared by P21 and Adamax — the second may not buy anything the first has not already bought. The real pairing logic is TAK-653: it is activity-dependent, so it makes the plasticity the other three propose actually get used. Adding capacity that is never engaged is the usual failure of a neurotrophic stack.
What we don't know
Whether this combination raises BDNF in humans at all is inferred, not demonstrated — the AMPA-to-BDNF link is a longer chain than the title suggests. P21's and Adamax's human effects cannot be predicted with confidence from their literature.
Reality check
TAK-653 is the only item with a plausible same-week effect, reported as clarity under load rather than stimulation — and a compound that does nothing at rest is easy to wrongly conclude does nothing at all. Several of TAK-653's papers are industry-authored, disclosed rather than hidden. NSI-189 and P21 are slow, with an honest possibility of nothing.
Stoic Cookie verdict
Rename it in your head to 'neurotrophic signaling, discussed' — the BDNF label is doing marketing work the evidence does not. As a discussion set it is coherent; as a promise of higher BDNF it is not.
